摘要
目的观察痛宁胶囊对偏头痛大鼠血浆一氧化氮(NO)和5-羟色胺(5-HT)浓度的影响,探讨痛宁胶囊治疗偏头痛的神经学机制。方法 SD大鼠随机分成6组:空白对照组、模型对照组、阳性药物对照组、研究药物组(痛宁胶囊高、中、低剂量组)。除空白对照组外,其余各组采用皮下注射硝酸甘油法复制大鼠偏头痛模型。30min后研究药物组给予痛宁胶囊高、中、低剂量药液灌胃。阳性药物对照组按照6mg/kg给予琥珀酸舒马普坦溶液灌胃。空白对照组和模型对照组予以等容积的蒸馏水灌胃。造模4h后腹主动脉采血,NO采用硝酸还原酶法测定,5-HT采用酶联免疫吸附法(ELISA)测定。结果与模型对照组比较,阳性药物对照组及痛宁胶囊高、中剂量组均能显著降低偏头痛大鼠血浆NO的浓度,且能明显升高血浆5-HT的浓度(P<0.05)。与阳性药物对照组比较,高剂量组降低NO浓度及升高5-HT浓度无统计学意义(P>0.05)。结论痛宁胶囊可以调节偏头痛大鼠血管活性物质和神经递质的水平,可能是痛宁胶囊治疗偏头痛的作用机制之一。
Objective To investigate the effects of Tongning capsule on nitric oxide(NO) and hydroxytamine(5-HT) in rats with nitroglycerin migraines.Methods Sprague-Dawley rats were randomly assigned to 6 groups:Blank control group,model control group,masculine drug control group,study drug groups(including high dose subgroup,middle dose subgroup and low dose subgroup).All rats(except blank control group) were reproduced the migraine models by subcutaneously administration of nitroglycerin.Masculine drug control group was administrated sumatriptan succinate physic liquor following the dose of 6 mg/kg.Blank control group and model control group were administrated corresponding volume of distilled water.After the migraine animal model were established for 4 h,plasma was collected from abdominal aorta.The concentration of NO was measured by nitrate reductase method and the concentration of 5-HT was measured by enzyme linked immunosorbent assay.Results Compared with model control group,the concentrations of NO in masculine drug control group,Tongning capsule high dose subgroup and middle dose subgroup were reduced,and the concentrations of 5-HT were increased(P0.05).There were no significances in concentration of NO and 5-HT between Tongning capsule high does subgroup and masculine drug control group(P0.05).Conclusion Tongning capsule could regulate the levels of vasoactive substances and neurotransmitters in migraine rats during the migraine attacks.This might be one of the possible mechanisms of Tongning capsule for the treatment of migraine.
