摘要
目的研究芎芷地龙汤对偏头痛模型体内NO、NOS的影响。方法健康雄性SD大鼠随机为4组:生理盐水组、模型组、舒马普坦组、中药治疗组。皮下注射NTG注射剂复制实验性偏头痛动物模型,经药物干预后在4小时、6小时两个时间点测定各组血液和脑干中NO、NOS的含量。结果(1)模型组血浆和组织中NO含量升高,且4小时高于6小时,两个时间点的含量与生理盐水组比较均有显著性差异(P<0.01);中药组和舒马普坦组血浆和组织中NO含量降低,两个时间点的含量与模型组比较均有显著性差异(P<0.05)。(2)模型组血浆和组织中NOS含量升高,且4小时高于6小时,两个时间点的含量与生理盐水组比较有显著性差异(P<0.05);中药组和舒马普坦组血浆和组织中NOS含量降低,两个时间点的含量与模型组比较有显著性差异(P<0.05)。结论芎芷地龙汤可以下调由硝酸甘油诱导的NO、NOS异常升高的水平,这种作用可以遏制偏头痛发病过程中一系列级联反应的恶性循环,从而发挥抗偏头痛的作用。
objective To study the effect of Xiongzhidilong-Decoction NO and NOS in migraine model. Methods healthy male SD rats were randomly divided into 4 groups : normal sodium control group, model control group, Sumatriptan control group, Xiongzhidilong-Decoction group. Reproduce migraine animal model with subcutaneous injection NTG, contents of NO and NOS in blood and brain stem were deter-mined after medicine interfered 4h and 6h. Results ( 1 ) The contents of NO in plasma and tissue of model group is heightened , which is significant compared with normal sodium group at 4h and 6h (P 〈 0. 01 ), the contents at 4h is higher than the contents at 6h. The contents of NO in plasma and tissue of Xiongzhidilong- Decoction group and Sumatriptan group were cut down, which is significant compared with model group at 4h and 6h (P 〈 0.05 ). (2)The contents of NOS in plasma and tissue of model group is heightened , which is significant compared with normal sodium group at 4h and 6h ( P 〈 0. 05 ) , the contents at 4h is higher than the contents at 6h. The contents of NOS in plasma and tissue of Xiongzhidilong-Deeoction group and Sumatriptan group were cut down, which is significant compared with model group at 4h and 6h (P 〈 0. 05 ). Conclusion Xiongzhidilong-Deeoction can down regulation the abnormally increased contents of NO and NOS which was induced by nitroglycerin. This effect can inhibit a series of cascade reaction in vicious cycle in migraine occurrence, maintain a organism equilibrium level all the time, play relieving pain role.
