摘要
建立高效液相色谱(HPLC)测定石斛兰中内源激素GA3、IAA、ABA的检测方法。石斛兰植株经80%冷甲醇匀浆提取,PVPP及C18 SPE小柱净化,调整酸度后经乙酸乙酯萃取,氮气吹干后用甲醇溶解。以甲醇∶乙腈∶磷酸盐缓冲液(体积比=15∶15∶70,pH=3.5)为流动相,采用Waters nova-pakC18柱分离,UV210 nm检测,外标法定量。结果表明,方法的平均回收率为84.2%~95.4%,RSD为4.7%~5.8%,检出限为0.02~0.06 mg/kg,整个提取分离、分析测定流程简便、快速、准确。
A determination method for the internal hormones(GA3,IAA ABA) in dendrobium by high performance liquid chromatography(HPLC) was developed.The dendrobium plant was homogenized in cold 80% methanol,purified by PVPP and cleanert C18 SPE,and extracted by ethyl acetate after the acidity regulated,then dissolved in methanol when the solvent was blown away by nitrogen.The internal hormones were seperated by Waters nova-pak C18 column,and determined by UV detector with the wavelength at 210 nm.The mobile phase was methanol∶acetonitrile∶phosphate(15∶15∶70,pH=3.5).The recovery range was 84.2% to 95.4% with RSD from 4.7% to 5.8%.The limits of the method varied from 0.02~0.06 mg/kg.It was proved that this procedure was simple,rapid and accurate for efficient determination of internal hormones in dendrobium plant.
