摘要
目的探讨牛磺酸对高温后神经上皮细胞存活的影响。方法取孕12 d小鼠胚胎神经管上皮细胞进行原代培养,将其分为正常对照组、高温对照组和5个牛磺酸组(浓度分别为2、6、10、15和20 mM)。通过MTT法测定细胞光密度值(OD值)以反应细胞增殖活性,DAPI染色法检测细胞凋亡。结果高温对照组细胞的增殖活性明显低于正常对照组(P<0.01),细胞凋亡明显增多;61、0 mM牛磺酸组细胞的增殖活性明显高于高温对照组(P<0.05),高温12 h后细胞凋亡率低于高温对照组(P<0.01),并持续至高温后48 h。结论 6 mM和10 mM牛磺酸对高温后神经上皮细胞的存活具有保护作用。
Objective To study the protective function of taurine in hyperthermia induced apoptosis in cultured neuroepithelial cells.Methods Neuroepithelial cells were dissociated from embryonic neural tube of E12d mice.They were divided into the control group,the hyperthermia group and 5 taurine groups of 2 mM,6 mM,10 mM,15 mM and 20 mM.Cell viability and apoptosis were determined by MTT assay and DAPI staining.Results In the hyperthermia group,compared to the control group,the proliferative activities of neuroepithelial cells decreased significantly(P0.01),the apoptosis percentage increased.The cell viability of 6 mM and 10 mM taurine groups was obviously higher than that of the hyperthermia group(P0.05).The apoptosis rates in 6mM and 10 mM taurine groups decreased 12 hours after hyperthermia compared with that of hyperthermia group(P0.01).Conclusion 6 mM and 10 mM of Taurine may protect neuroepithelial cells from hyperthermia.
