摘要
目的研究大鼠创伤性脑损伤后血脑屏障(BBB)通透性的变化。方法参照Feeney’s自由落体致伤法,制作大鼠顶叶局灶皮质挫裂伤模型。应用伊文氏兰(Evans Blue,EB)示踪,测定伤后不同时点皮层和海马脑组织中EB含量;应用荧光显微镜观察皮层和海马是否存在EB的渗出。结果损伤灶皮层和同侧海马中EB含量在伤后1~6h与伤后1~7d明显增多,高峰期在3h和3d。荧光显微镜下发现血脑屏障中间期损伤灶同侧海马仍然存在EB的渗出。结论大鼠顶叶局部脑挫裂伤后,损伤灶皮层和同侧海马BBB开放呈双相性变化,BBB开放中间期仍然有很少量EB渗出。
Objective To study the changes of permeability of blood brain barrier(BBB) after traumatic brain injury in rats.Methods Rats were randomly divided into control group and brain injury group.Brain injury animal modes were established according to Feeney's method.Measurement of Evans blue(EB) in both cortex and hippocampus was performed at different time point after injury,and the EB extravasation was observed by fluorescence microscope.Results The content of EB in ipsilateral cortex and hippocampus was increased obviously at the time point of 1 to 6 h and 1 to 7 d after injury.The extravasation peak course was 3h and 3d after injury.Under fluorescence microscope,red fluorescence was observed in ipsilateral hippocampus 12h after injury.Conclusion After traumatic brain injury in rats,the BBB opening in ipsilateral cortex and ipsilateral hippocampus is biphasic.Zero-leakage-rate permeability is not exit between the two opening period.
