抗郁散对慢性不可预见性轻度应激抑郁症模型大鼠海马5-HT_(1A)R基因表达的影响 被引量:1

Study on effects of Kangyusan on mRNA expression of 5-HT_(1A)R of hippocampus in CUMS rat

摘要 目的:观察抗郁散对慢性不可预见性轻度应激抑郁症模型(CUMS)大鼠的行为学变化及其大脑海马5-HT1AR基因表达水平的影响。方法:将46只成年雄性SD大鼠,随机分为正常空白组、模型对照组、丁螺环酮组和抗郁散组4组,除正常组外其余大鼠利用CUMS方法复制抑郁模型。运用RT-PCR技术扩增,凝胶图像分析系统中对各目的基因条带的浓度进行测定。结果:抗郁散组与模型组比较,大鼠的体质量明显增加(P<0.05),水平和垂直行为活动明显增多(P<0.05),对糖水消耗明显增强(P<0.05),海马5-HT1AR基因表达显著提高(P<0.01)。结论:抗郁散具有改善CUMS大鼠行为学变化的特征,明显提高CUMS大鼠海马5-HT1AR的mRNA表达水平,提高5-HT的活性从而达到治疗抑郁症的目的。 Objective:The present study was designed to investigate the effects of Kangyusan on mRNA gene expression of 5-Hydroxytryptamine 1A receptor in hippocampus and behavior in CUMS rat.Methods:46 adult male clean SD rats were resigned randomly into the control,model,buspirone and Kangyusan groups.The rats were exposed to CUMS for more than 21 days apart from control group.It was observed that rat's behaviors and determined that level of 5-HT1ARmRNA gene express of hippocampus in rats by RT-PCR amplification and the GeneGeniu Image Analysis System.Results:Compared with model group,the performence has showed that weight,crossing move,rearing move and sucrose consumption were incased significantly in Kangyusan group(P0.05,respectively);and more 5-HT1ARmRNA gene expression of hippocampus was increased significantly(P0.01).Conclusions:The experiments suggested that weight,behavior,sucrose consumption had been improved obviously in CUMS rats administrated with Kangyusan,and the level of 5-HT1ARmRNA gene expression were increased signicantly,which could demonstrate increasing 5-HT activity in the hippocampus of CUMS rats and it's useful in the treatment for depression.
