摘要
目的评价海妇洁栓对大鼠胚胎发育的影响。方法将Wistar妊娠大鼠随机分为5组:基质对照组、阳性对照(环磷酰胺7 mg/kg)组、海妇洁栓低(16 mg/kg)、中(32 mg/kg)和高(80 mg/kg)剂量组。基质对照组和海妇洁栓各剂量组孕鼠于妊娠第6天开始经阴道给药,每天1次,连续10 d;阳性对照组孕鼠于妊娠第11天开始经大腿外侧肌肉注射给药,每天1次,连续3 d。于妊娠第20天剖腹取出活胎,观察海妇洁栓对大鼠胚胎发育的影响。结果除阳性对照组外,海妇洁栓各剂量组及基质对照组所有受检胎鼠均未见明显外观畸形;平均体重、身长及尾长虽组间比较差异有统计学意义(P<0.05),但均属于正常生理变化,无生物学及毒理学意义,与海妇洁栓无关;除阳性对照组外,海妇洁栓各剂量组及基质对照组所有受检胎鼠骨骼、主要内脏器官均未见明显异常。结论海妇洁栓剂量在16~80 mg/kg之间对Wistar大鼠胚胎无明显的毒性和致畸作用。
Objective To evaluate the effect of marine lysozyme vaginal suppository on development of rat embryo.Methods Pregnant Wistar rats were randomly divided into matrix control,positive control and three test groups.The rats in three test groups were treated with marine lysozyme vaginal suppository at low(16 mg / kg),moderate(32 mg / kg) and high(80 mg / kg) dosages respectively starting from day 6 after pregnancy,once a day for 10 d,while those in matrix control group with lysozyme-free matrix suppository.However,the rats in positive control group were injected i.m.with cyclophosphamide at a dosage of 7 mg / kg starting from day 11 after pregnancy,once a day for 3 d.The rats were killed on day 20 after pregnancy,and live fetal rats were taken out to evaluate the effect of marine lysozyme vaginal suppository on development of rat embryo.Results No obvious deformities were observed in the fetal rats in various groups except positive group.Though the mean bodyweights,body lengths and tail lengths of fetal rats in various groups showed significant difference(P〈0.05),all the indexes were within the range of normal physiological change and showed no significance in biology or toxicology.Except those in positive control group,no obvious abnormalities were observed in bones or main organs of fetal rats in various groups.Conclusion Marine lysozyme vaginal suppository at a dosage range of 16 ~ 80 mg / kg showed no significant toxicity or teratogenicity to the embryos of Wistar rats.
