摘要
目的 :观察不同浓度去甲肾上腺素 ( NA,10 - 1 0 ~ 10 - 6mol/ L )对大鼠 B细胞功能的影响 ,α和β肾上腺素能受体及钙离子通道在 NA影响 B细胞功能中的作用 ,从细胞水平了解 NA对 B细胞功能的调节作用及其作用机制。方法 :利用体外抗体生成的检测方法 ,即用绵羊红细胞 ( SRBC)刺激大鼠肠系膜淋巴结 B细胞转化成抗体形成细胞 ( AFC) ,然后检测其抗体生成量。结果 :( 1)10 - 1 0、10 - 9、10 - 8和 10 - 7mol/ L NA都能显著抑制大鼠 B细胞功能 ,其中 10 - 8mol/ L NA的作用最强 ,而 10 - 6mol/ L NA无明显的抑制作用。( 2 )α受体拮抗剂酚妥拉明 ( phentolamine,10 - 7mol/ L )和β受体拮抗剂心得安 ( propranolol,10 - 7mol/ L )分别作用于淋巴细胞时 ,都不能阻断 NA抑制 B细胞功能的作用 ,而两者共同作用于淋巴细胞时 ,能完全阻断 NA抑制 B细胞功能的作用。 ( 3 )钙通道阻滞剂维拉帕米 ( verapamil,10 - 7mol/ L )和尼群地平 ( nitrendipine,10 - 7mol/ L )都能阻滞 NA抑制 B细胞功能的作用。结论 :NA可非浓度依赖性地抑制大鼠 B细胞功能 ,此作用可能由
Objective:Noradrenaline (NA) has been supposed to be one of the important neurotransmitters modulating immune function. In the present study, the effect of NA on the B-lymphocyte function and the primary mechanism were investigated. Methods: Various concentrations of NA (10 -10 to 10 -6 mol/L) or adrenerceptor antagonist or calcium antagonist with sheep red blood cells (SRBC) were simultaneously added to the lymphocyte suspension of mesenteric lymph node of rats and incubated five days. Then OD value to reflect the B-lymphocyte function was determined with the aid of the assay for antibody synthesis in vitro.Results: NA at the concentrations of 10 -10 , 10 -9 , 10 -8 or 10 -7 mol/L significantly suppressed the B-lymphocyte function, having the strongest suppression at 10 -8 mol/L. NA at 10 -6 mol/L did not cause remarkable decrease in the B-lymphocyte function. When α-adrenerceptor antagonist phentolamine or β-adrenerceptor antagonist propranolol respectively with NA acted on the lymphocytes, the antibody synthesis still significantly decreased, without remarkable difference from the action of NA alone. However, when phentolamine and propranolol acted together with NA on the lymphocytes, the inhibition of antibody synthesis by NA was completely blocked. The calcium antagonists verapamil or nitrendipine also blocked the inhibitory effect of NA on the antibody synthesis in vitro. Conclusion: NA can cause concentration-independent suppression of the B-lymphocyte function of rats in vitro, which may be mediated either by α or by β adrenergic receptors and may be related to calcium increase within B-lymphocytes.
