摘要
目的观察银杏叶总黄酮(TFG)对肾缺血-再灌注(I-R)损伤大鼠p38丝裂素活化蛋白激酶(MAPK)活性的影响。方法 120只SD大鼠随机均分为假手术组I、-R组和TFG预处理组(TFG 250 mg/kg,每天3次)。3 d后采用夹闭双侧肾蒂致肾缺血45 min后再灌注制作大鼠肾I-R损伤模型。Western blot检测p38 MAPK和磷酸化p38 MAPK蛋白表达;免疫组化方法检测肾组织磷酸化p38 MAPK的分布情况。结果磷酸化p38 MAPK表达活性在再灌注10 min开始增加,1 h达最高峰,持续至24 h仍高于正常水平,且主要在肾小管细胞表达。TFG预处理组磷酸化p38 MAPK的表达较I-R组明显减弱(P<0.01)。结论 TFG可能通过抑制肾I-R损伤大鼠p38 MAPK的活化而发挥防治作用。
Objective To observe the effect of total flavonoid in leaves of Ginkgo biloba(TFG) on expression of p38 mitogen-activated protein kinase(MAPK) after renal ischemia-reperfusion(I-R) in rats.Methods One hundred and twenty rats were randomly divided into sham operation group,I-R group and TFG pretreatment group(TFG 250mg/kg,3 times a day) with 40 rats each.After 3 days the rat model of renal I-R injury was established by clamping renal pedicles to induce kidney ischemia for 45min and reperfusion thenafter.The expressions of p38 MAPK and phospho-p38 MAPK protein were examined by Western blot.The distribution of phospho-p38 MAPK in renal tissue was assayed by immunohistochemistry.Results The activation of phospho-p38 MAPK was markedly increased at 10min after reperfusion,reached its peak at 1h and continued to 24h,which was mainly located in renal tubular cells.Compared with I-R group,the expression of phospho-p38 MAPK was markedly reduced in TFG pretreatment group(P〈0.01).Conclusion TFG is able to protect the kidney from I-R injury through inhibiting p38 MAPK activation.
