摘要
目的:寻找一种简便有效测定壳寡糖的方法。方法:采用Elson-Morgan法、苯酚-硫酸法和3,5-二硝基水杨酸(DNS)法测定壳寡糖含量并进行比较。结果:Elson-Morgan法不适用于壳寡糖含量的测定;苯酚-硫酸法中盐酸氨基葡萄糖在200~3000μg范围与吸光度也有很好的线性关系,但苯酚-硫酸法对壳寡糖的测定值较实际值偏大;DNS法测定壳寡糖与实际值接近,平均回收率为96.1%,RSD为2.9%(n=6)。结论:DNS法操作简单、快速,重现性好,可直接用于壳寡糖含量的测定。
Objective:To find a simple and effective method for determination of chitooligosaccharide.Method:Elson-Morgan method,phenol-sulphuric acid method and 3,5-dinitrosalicyacid method(DNS) method were employed to determine content of chitooligosaccharide by UV spectrophotography.Result:Elson-Morgan method can not be used for determination of chitooligosaccharide;Glucosamine hydrochloride can be reacted with phenol-sulphuric acid and was linear with absorbency in 200-3000 μg,but the determination value was higher than the actual value by phenol-sulphuric acid method;The result of DNS method was more reliable and accurate,the average recovery ratio was 96.1%,RSD=2.9%(n=6).Conclusion:DNS method is simple,fast,repeatable and suitable for the determination content of chitooligosaccharide in solution.
