肌纤维母细胞在癌与非癌组织内差异性蛋白表达 被引量:1

摘要 目的筛选出癌相关肌纤维母细胞(carcinoma associated myofibroblasts,CAMs),与良性病变中增生的肌纤维母细胞(normal myofibroblasts,NMs)之间不同的差异性蛋白表达。方法将肌纤维母细胞增生病例73例分为癌与非癌组。癌组40例(54.8%),非癌组33例(45.2%),采用EnVision免疫组织化学法分别检测6个肿瘤间质表达蛋白:基质细胞衍生因子(stromal cell derived factor-1,SDF-1)、富于半胱氨酸酸性分泌蛋白(secreted protein,acidic and rich in cysteine,SPARC)、小窝蛋白1(caveolin-1,Cav-1)、纤维母细胞生长因子-2(fibroblast growth factor-2,FGF-2)、腱糖蛋白C(tenascin C,TNC)和成骨细胞特异性因子2(osteoblast-specific factor 2,OSF-2)的表达,分析非癌组与癌间质内差异具有统计学意义表达的蛋白分子。以平滑肌肌动蛋白(smooth-muscle actin,α-SMA)及血小板衍生生长因子(platelet-derived growth factor-βreceptor,PDGFR-β)作为鉴定肌纤维母细胞的特异性标志物。结果 6个检测指标中仅有Cav-1与α-SMA/PDGFR-β的阳性表达部位最为相似;Cav-1在间质内表达的阳性率,癌组为85.0%(34/40)显著高于非癌组45.5%(15/33),差异具有统计学意义(P<0.05)。SPARC和TNC在癌组与非癌组阳性率均达到100.0%,按阳性程度划分为高表达与低表达两组后,非癌组内间质的表达水平显著高于癌组,且差异具有统计学意义(P<0.05)。结论癌与非癌组织内的肌纤维母细胞在Cav-1、SPARC和TNC的蛋白质表达水平上有差异性。Cav-1癌组表达水平显著高于非癌组,且与CAMs标志物α-SMA的阳性表达部位最为相似,可作为CAMs的定性辅助检测指标。 Objective To investigate the differential protein expression in carcinoma-associated myofibroblasts(CAMs) and normal myofibroblasts(NMs). Methods Seventy-three cases with desmoplastic reaction were divided as cancer(n=40) and non-cancer(n=33) groups.The expression of 6 proteins,including stromal cell derived factor-1(SDF-1),secreted protein acidic and rich in cysteine(SPARC),caveolin-1(Cav-1),fibroblast growth factor-2(FGF2),tenascin C(TNC) and osteoblast specific factor 2(OSF-2) was detected by immunohistochemistry EnVision method in tissue specimens of two groups.CAMs was defined by the expression of α-SMA and PDGFR-β receptors. Results Among these biomarkers,only CAV-1 showed a close approximation of immunoreactivity to α-SMA-positive cells.Positive rate of stromal Cav-1 expression in cancer group was 85.0%(34/40),significant higher than that in non-cancer group(45.5%,15/33)(P0.05).The positive rates of SPARC and TNC expression were 100.0% in both groups,but the intensity of SPARC and TNC expression in NMs group was significantly higher than that in CAMs group(P0.05). Conclusion There are differential expressions of Cav-1,SPARC and TNC proteins between CAMs and normal myofibroblasts.The expression pattern of Cav-1 in CAMs is similar to SMA,which suggests that Cav-1 might be used as a marker for CAMs.
