摘要 目的:探讨白血病患者骨髓中CD105、VEGF及bFGF的表达及临床意义。方法:采用原位杂交方法,用CD105标记骨髓活化的血管内皮细胞;用ELISA方法检测骨髓液VEGF、bFGF的表达水平。结果:CD105在白血病的表达以急性髓系白血病(AML-M5)最高,急性髓系白血病(AML)AML-M3表达最低。AML-M3、CLL的阳性表达率与正常对照组差异无统计学意义(P>0.05);其他白血病的阳性表达率均明显高于正常对照组(P<0.05);骨髓液VEGF含量在CML表达最高,bFGF在B-ALL表达最高。在CLL中,VEGF、bFGF含量与正常对照组差异无统计学意义(P>0.05);在T-ALL中,bFGF含量与正常对照组差异无统计学意义(P>0.05),VEGF含量明显高于正常对照组(P<0.05);其他白血病VEGF、bFGF均明显高于正常对照组(P<0.05);骨髓液CD105在AML-3的表达与VEGF、bFGF的表达呈负相关。在其他类型白血病与VEGF、bFGF的表达呈正相关。结论:白血病患者骨髓中血管新生因子存在异常表达,在白血病细胞的生长、侵润、转移过程中具有重要作用。 Objective To investigate the clinical significance of CD105, VEGF and bFGF expressions in bone marrow of the patients with leukemia. Methods The VEGF, bFGF expression levels in bone marrow were detected by means of ELISA, and the activated vascular endothelial cells in bone marrow were marked by CD105 using hybridization in situ. Results The expression level of CD105 was the highest in the patients with acute myeloid leukemia (AML-MS), and the lowest in the patients with acute myeloid leukemia (AML-M3). The positive rate of AML-M3 expression, CLL masculine had no significant difference in comparison with the control group (P 〉 0.05). The positive expression rates of other leukemia were significantly higher than normal control group (P 〈 0.05). VEGF content was the highest in bone marrow of CML, bFGF was the highest in B-ALL. VEGF, bFGF content in CLL had no statistically significant difference with normal control group (P 〉 0.05 ). BFGF content in T- ALL had no statistically significant difference with normal control group (P 〉 0.05). There was statistically significant difference in VEGF content between T-ALL and normal control group (P 〈 0.05).VEGF contents in other leukemia were significantly higher than that in normal control group(P 〈 0.05). CD105 level in bone marrow liquid of AML-3 was negatively correlated with VEGF, bFGF expressions, but it was positively correlated in other types of leukemia. Conclusion The angiogenic factor expressions are unnormal in bone marrow of the patients with leukemia, and paly an important role in the growth, invasion, and metastasis of leukemia.
