家兔耳轻度冻伤模型构建中浸冻时间的筛选研究 被引量:3

Selection of Freezing Duration in the Establishment of Slight Frostbite Model of Rabbit Ear

摘要 目的:确立构建家兔耳轻度冻伤模型的浸冻时间。方法:将家兔随机分为正常对照组、浸冻20s组和浸冻30s组,每组6只,将后2组兔耳同时浸入-24.5℃、95%乙醇中冻结相应时间,观察8d内各组兔耳的外观、组织病理学及肿胀度变化。结果:浸冻20s组兔耳冻后12~48h有小水泡,冻后8d受冻部位基本恢复正常,镜下可见表皮增厚,皮肤附件和软骨组织均未见明显变化;浸冻30s组兔耳冻后12~48h出现浆液性水泡,冻后8d受冻部位仍有水肿,并伴随皮肤脱落,镜下可见表皮溃破、缺损,皮肤附件和软骨组织周围有炎性细胞浸润;从冻后3d起,浸冻30s组兔耳的肿胀度明显高于浸冻20s组(P<0.05)。结论:可通过在-24.5℃、95%乙醇浴中浸冻20s构建家兔耳轻度冻伤模型。 OBJECTIVE : To explore the freezing duration of slight frostbite model of rabbit ear. METHODS: Rabbis were ran- domly divided into normal control group, 20 s freezing group and 30 s freezing group with 6 rabbits in each group. Rabbit ears were soaked in 95% alcoholic balneum frigidum at -- 24.5 ℃ for different time (20 s and 30 s), then the appearance of rabbit ears, histopathology, swelling and distension degree of them in 8 d were observed. RESULTS: After freezing for 20 s, the skin of rabbit ear changed into small blister 12--48 h after freezing, and the injured site turned into normal 8 d after freezing; the thickening of skin could be observed and appendant of skin and cartilaginous tissue had no obvious change. After freezing for 30 s, the rabbit ear changed into serous blister 12--48 h after freezing and the injured site appeared edema after 8 d and the skin began to shedding; epidermis diabrosis and lesion could be observed under microscope and appendant of skin and cartilaginous tissue surround were infiltrated with inflammatory cells. Since 3 d after freezing, the swelling and distension degree of rabbit ears in 30 s freezing group was significantly higher than in 20 s freezing group (P〈0.05). CONCLUSION: Slight frostbite model of rabbit ear can be established by freezing them in 95% alcoholic balneum frigidum at --24.5 ℃ for 20 s.
