摘要
为探讨口腔癌组织中KLF6、p21和c-Jun的表达及意义,采用免疫组化技术检测198例口腔癌、49例口腔良性肿瘤中KLF6、p21和c-Jun的表达,以56例口腔正常组织作对照。结果显示:正常对照组、良性及恶性肿瘤KLF6的表达分别为91.07%(51/56)、83.67%(41/49)、62.12%(123/198),三组间阳性表达有显著性差异(P<0.05);KLF6在肿瘤直径>2 cm组和中、低分化组中的阳性表达显著低于肿瘤直径≤2 cm组及高分化组(P<0.01);p21在恶性肿瘤中阳性表达81.82%(162/198)显著低于正常组的85.71%(48/56)(P<0.05);c-Jun在正常对照组、良性肿瘤及恶性肿瘤的表达分别为8.93%(5/56)、10.20%(5/49)、46.97%(93/198),三组间阳性表达有显著性差异(P<0.05)。恶性肿瘤中KLF6基因核表达缺失或减弱与p21表达下调显著相关(P<0.01),恶性肿瘤中KLF6浆表达增加与p21表达下调显著相关(P<0.01)。恶性肿瘤中KLF6核表达缺失或减弱与c-Jun阳性表达增加显著相关(P<0.01)。可见,KLF6、p21和c-Jun三者表达异常与口腔癌的发生发展密切相关,其分子机制值得深入研究。
To assess the expression of KLF6,p21 and c-Jun in oral squamous cell carcinoma,the expression of KLF6,p21 and c-Jun were detected by immunohistochemistry among 198 cases of cancerous tissue,49 cases of hyperplastic tissue,compared with 56 controls of normal tissue. The expression of KLF6 in the group of normal,hyperplastic and cancerous tissue were 91. 07%(51/56), 83. 67%(41/49), 62.12% (123/198),respectively. There were significant differences among the three groups (P〈0.05). The positive expression of KLF6 in the group of tumor〉2 cm and moderate differentiated and poorly differentiated were significantly lower than that in the group with tumor〉2 cm and high differentiated (P〈0.01). The positive expression of p21 in cancerous tissue of 81.82%(162/198) was significantly lower than that in normal tissue with 85.71%(48/56) (PM0.05). The expression of c-Jun in the group of normal,hyperplastic and cancerous tissue were 8.93%(5/56) ,10. 20%(5/49) ,46.97%(93/198) ,respectively. There were significant differences among the three groups (P〈0.05). The deletion or reducing of KLF6 expression in nucleus and the increasing of KLF6 expression in cytoplasm were significantly correlated with the downregulation of p21 expression in the cancerous tissues (P〈0. 05). The deletion or reducing of KLF6 expression in nucleus was significantly correlated with the increasing of c-Jun expression in the cancerous tissues (P〈0.01). The results indicate that the abnormal expression of KLF6, p21 and c-Jun may involve in the development of the oral squamous cell carcinoma. Further research should be done to find out the mechanism and correlativity.
