狭叶红景天中槲皮素和木犀草素含量测定方法的建立 被引量:3

摘要 目的为控制狭叶红景天的质量,建立狭叶红景天中槲皮素和木犀草素含量的反相高效液相色谱测定法。方法采用Hypersil ODS-2(5μm,4.6×250mm)色谱柱,以甲醇:水(含0.05%的磷酸)=48:52为流动相进行洗脱,流速为1 mL.min-1,检测波长为360 nm,柱温35℃,外标法测定狭叶红景天中槲皮素和木犀草素的含量。结果槲皮素和木犀草素与狭叶红景天中所含的其他物质均达到基线分离,线性范围分别为0.14~0.7μg(r=0.9997),0.168~0.84μg(r=0.9999);平均回收率为100.34%(RSD=2.83%)、99.79%(RSD=2.04%)。结论所建方法测定快速,结果准确、可靠,可以用于狭叶红景天中槲皮素和木犀草素含量的测定。 Objective To establish a HPLC method for determination of quercetin and luteolin in Rhodiola kirilowii.Methods Hypersil ODS-2(4.6mm×250mm,5μm) column was used to determine the contents of quercetin and luteolin under the condition of mobile phase of methanol:water(0.05% phosphoric acid) = 48:52,flow rate of 1mL/ min and detection wavelength of 360nm at 35℃.Results The quercetin,luteolin and other substances in Rhodiola kirilowii were reached at baseline separation.The linear ranges of quercetin and luteolin was 0.14~0.7 μg(r=0.9997) and 0.168~0.84 μg(r=0.9999),respectively.The average recoveries was 100.34 %(RSD=2.83%) and 99.79 %(RSD=2.04%).Conclusion This method is rapid,accurate,reliable and can be used in detection of the contents of quercetin and luteolin in Rhodiola kirilowii.
