摘要
目的探讨白藜芦醇对非酒精性脂肪肝大鼠的分子机制。方法实验分组:正常对照组;模型组;白藜芦醇低剂量组;白藜芦醇高剂量组。正常SD大鼠高脂饲养8周,复制大鼠非酒精性脂肪肝模型,组织病理学观察,Real-time PCR检测氧化及凋亡相关基因,血清生化检测。结果白藜芦醇治疗组和模型组比较,形态学显示治疗组肝肌细胞变性坏死明显减轻;氧化应激相关指标SOD1活性增高;均有显著性差异(P<0.05)。结论白藜芦醇调控氧化应激相关基因表达,减轻肝细胞的脂质过氧化损伤,改善肝脏功能。
OBJECTIVE To investigate the molecular mechanism of resveratrol's effect on rats with non-alcoholic fatty liver.METHODS We established normal control group,model group,low dosage resveratrol group,and high dosage resveratrol group.The model of non-alcoholic fatty liver from normal SD rats was duplicated after high fat diet was administrated for 8 weeks.Histopathologic observation,biochemical test for serum,and real time PCR for detecting the related genes of oxidation and apoptosis were carried out.RESULTS Compared with model group,denaturation and necrosis of hepatocytes in resveratrol group were slighter,and SOD1 increased,showing significant differences(P0.05).CONCLUSION Resveratrol adjusts and controls the expression of related genes of oxidative stress,and alleviates lipid peroxidation impairment of hepatic cells to improve liver's function.
