摘要
目的:观察大黄酸对肥胖糖尿病大鼠肾组织氧化应激反应的影响。方法:高脂膳食加链脲佐菌素诱导的肥胖糖尿病大鼠给予大黄酸治疗8周后,检测大黄酸对大鼠血糖、24 h尿蛋白排泄量及肾皮质超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量等的影响。结果:与正常对照组相比,糖尿病大鼠肾皮质MDA含量增加,SOD活性明显降低,同时伴尿蛋白排泄量升高;大黄酸治疗可使肾组织MDA含量降低,SOD活性显著增加,血糖和体重也有明显下降。结论:糖尿病大鼠肾组织存在明显的氧化应激反应,大黄酸具有的抗氧化活性可能是其防治糖尿病肾病的机制之一。
Objective:To study the effect of Rhein on oxidative stress of the kidneys in fat diabetic rats.Methods:The fat diabetic rats were induced by streptozotocin and high fat diet.The rats were divided into 3 groups by random:control group(NC),diabetes mellitus group(DM)and diabetic rats receiving Rhein(DM+RH).Rhein was given at a dose of 100mg·kg-1·d-1 once daily.After administration by gavage for 8 weeks,blood glucose,body weight,24h urinary protein excretion,the levels of malondialdehyde(MDA),total activities of superoxide dismutase(T-SOD) were measured.Results:Elevated MDA level in diabetic renal tissue decreased and the levels of activities of T-SOD in diabetic renal tissue were also remitted by Rhein(P0.05,P0.01).24h urinary protein excretion were significantly attenuated by treatment with Rhein(P0.05),and Rhein reduced the level of plasma glucose in diabetic rats(P0.05).Conclusion:This study shows that the kidneys of fat diabetic rats undergo an obvious oxidative stress,and antioxidation may be one of the possible mechanisms that Rhein can protect and remitte diabetic nephropathy.
