消瘀泄浊饮对肾间质纤维化大鼠的肾脏保护作用研究 被引量:17

摘要 目的:探讨消瘀泄浊饮对UUO模型大鼠的保护作用及作用机制。方法:实验采用UUO大鼠模型,将32只SD大鼠随机分为假手术组(S组)、模型组(M组)、洛汀新治疗组(B组)、消瘀泄浊饮组(X组)。分组用药并监测体重,给药14天后处死大鼠。测定大鼠左肾指数,血清肌酐、尿素氮、白蛋白水平;肾组织HE染色观察病理改变;用免疫组织化学方法检测肾组织α-SMA的表达。结果:消瘀泄浊饮能够明显降低实验大鼠血清肌酐、尿素氮水平,升高白蛋白水平,能降低肾组织中α-SMA蛋白表达,与模型组比较有显著性差异,形态学观察也显示其肾脏损害轻于模型组。结论:消瘀泄浊饮能有效地延缓肾间质纤维化的进程,可通过抑制α-SMA的表达,减少肌成纤维细胞的活化和形成,从而起到抗肾纤维化的作用。 Objective:To investigate the protective effect and mechanism of Xiaoyu Xiezhuo Yin on UUO rats.Methods:In the experiment,unilateral ureteric obstruction(UUO) model is adopted,32 SD rats were randomly divided into 4 groups:sham-operated group(S group),model group(M group),benazepril treated group(B group),Xiaoyu Xiezhuo Yin treated group(X group).After being intragastric administrated respectively and monitored the weight for 2 weeks,all rats were executed.Left-kidney index,levels of serum creatinine,urea nitrogen,albumin were detected;nephridial tissue were stained by HE to observe the pathologic change;immunohistochemistry was applied to measure the expression of α-SMA.Results:Xiaoyu Xiezhuo Yin decreased the level of serum creatinine,urea nitrogen and increased the the level of serum albumin obviously,and it reduced the expression of α-SMA protein in nephridial tissue,which has the significant difference compared with M group,morever,pathological changes in the X group were milder than those in the M group.Conclusion:Xiaoyu Xiezhuo Yin can effecticely delay the progression of renal fibrosis.Morever it can inhibition the expression of α-SMA and decrease the activation and formation of myofibroblasts,which is the main mechnism of anti-fibrosis.
