摘要
目的用血清药理学的方法研究宣导泻肺饮含药血清对大鼠心室肌细胞L-型钙通道的影响。方法采用全细胞膜片钳技术记录L-型钙离子通道电流(Ica-L),观察不同浓度的宣导泻肺饮对它们的影响。结果宣导泻肺饮含药血清对心肌细胞L-型钙离子通道的开放有抑制作用,在浓度10%时Ica-L峰值降低15%,浓度30%时Ica-L峰值降42%,均呈浓度依赖性;随着药物浓度的增加,电流-电压关系曲线逐渐上移,但出现电流峰值的电压不变。结论宣导泻肺饮含药血清抑制心肌细胞钙离子通道的开放,具有明显的浓度依赖关系,可能是其抗心律失常的分子基础。
Objective:To study the effects of Xuandaoxiefei Decoction on L-type calcium channel in ventricular myocyte.Methods:The L-type calcium current(Ica-L) was recorded by the whole-cell patch clamp technique to observe the effect of Xuandaoxiefei Decoction with difference concentrations.Results:The of L-type calcium channel was inhibited by the Xuandaoxiefei Decoction.The peak of Ica-L was reduced 15% by the Xuandaoxiefei Decoction concentration with 10%.And the peak was reduced 42% by the concentration with 30%.Above peak indices were in the concentration-dependent manner.As increase of the decoction level,the current-voltage curve lift gradually.However,the voltage was invariant when the current peak existed.Conclusion:That the opening of Ica-L in ventricular myocyte is inhibit by the Xuandaoxiefei Decoction with the obvious concentration-dependent manner could be the molecular basis of anti-arryhthmia.
