摘要
目的优选本院制剂胆胰宁合剂澄清工艺。方法采用单因素试验方法,以黄芩苷含量、澄清度及干膏率为评价指标,对醇沉法、调pH醇沉法、ZTC絮凝法、壳聚糖絮凝法和离心法进行研究,筛选出适宜的澄清方法;并对药液的pH值、醇沉浓度、醇沉温度进行考察,优化胆胰宁合剂澄清工艺。结果适宜的澄清方法是调pH醇沉法,优化的澄清工艺为:药液调pH值至6,醇沉浓度60%,醇沉温度60℃。结论调pH醇沉法能有效除去杂质,保留有效成分,操作简便,可作为胆胰宁合剂的澄清工艺。
Objective To optimize the clarification process of hospital preparation Danyining mixture.Methods Danyining mixture was clarified respectively with ethanol precipitation,ethanol precipitation with adjusting pH,centrifugation,chitosan flocculation and ZTC flocculation,the suitable method was determined.Based on the method,the optimum clarification conditions were investigated by single factor experiments with the content of baicalin,rate of dried extract and clarity as indexes.Results The appropriate clarification method was ethanol precipitation with adjusting pH.The best technological parameters were as follows：at ethanol concentration of 60%,pH of 6,precipitation temperature of 60 ℃.Conclusion The ethanol precipitation with adjusting pH can remove impurities effectively,keep active ingredients and simplify operation,so it can be used as clarification technology of hospital preparation Danyining mixture.
