摘要
苯巴比妥钠诱导增殖的小鼠肝脏加热提取物5 ~200mg/L,明显刺激人L02 肝细胞系3 H- TdR掺入,抑制5 - Fu 对人L02 肝细胞的杀伤作用; 相同剂量的非加热提取物抑制人L02 肝细胞生长。苯巴比妥钠5 ~20mg/L,对人L02 细胞3 H- TdR 掺入及5 - Fu 杀伤作用无影响。经胰蛋白酶预处理后, 小鼠增殖肝加热提取物活性消失。这说明苯巴比妥钠本身对人L02 肝细胞不是一种丝裂原性药物, 其诱导小鼠肝脏增殖作用与肝脏局部体液因素有关, 该因素为蛋白或肽类物质。
The heated extract from mouse hyperplastic liver induced by phenobarbital sodium (5~200mg/L) significantly stimulated the incorporation of 3H-TdR in human L 02 liver cell line, and inhibited the killing of 5-Fu on the cell line. The similar dose of the unheated extract inhibited the growth of the L 02 cell. Phenobarbital sodium 5~20mg/L did not affect the incorporation of 3H-TdR in the L 02 cell line and the killing action of 5-Fu on it. The activity of the heated extract was eliminated by the pretreatment with trypsin. The results showed that phenobarbital itself was not a mitogenic drug in the L 02 cell culture and the induction of hepatic proliferation by phenobarbital was related to local humoral factor in mouse liver, which was an intrinsic protein or peptide substance.
