摘要
目的 研究不同浓度的小牛肝提取物对新生小鼠肝细胞损伤的影响。方法 采用Ⅳ型胶原酶消化法制备游离肝细胞悬液,以低血清进行肝细胞原代培养,12-16 h后用0.1 mmol/L的H2O2诱导肝细胞损伤,同时加以不同浓度的肝提取物,观察(100-1 000)mg/L的小牛肝提取物对肝细胞损伤的影响。结果 小牛肝提取物在100 mg/L时即可起到保护肝细胞损伤的作用,但效果不明显,在500 mg/L时可明显保护肝细胞损伤,之后随着浓度的加大保护作用逐渐减弱。结论 小牛肝提取物对H2O2诱导的肝细胞损伤具有一定的保护作用,其中以浓度为500 mg/L时效果最明显。
Objective To study the effect of different concentrations of calf liver extract on the damage of the neonatal mice hepatocytes. Methods Hepatocytes were digested by IV - Collagenase and cultured with low - serum medium, induced by H2O2 after 12 - 16 h. Various concentrations of calf liver extract were added to the primary, cultured hepatocytes to observe the effect of ( 100 - 1 000) mg/L calf liver extract on the injured liver cell. Results Calf liver extract at 100 mg/L could protect the hepatocytes from injury,, but the effect was not obvious. At 500 mg/L, calf liver extract could protect the hepatocytes obviously. With increasing concentration, the protective effect weakened gradually. Conclusions Calf liver extract could protect hepatocytes from damage induced by H2O2 , the effect at a concentration of 500 mg/L was the most obvious.
