摘要
目的研究正常晚期妊娠以及子痫前期(preeclampsia,PE)胎盘组织中αvβ3基因和蛋白表达的差异,探讨αvβ3与子痫前期的关系。方法采用免疫组化及Western blot方法检测25例PE患者(其中轻度PE组10例,重度PE组15例)和20例正常足月孕妇(对照组)胎盘组织中αvβ3蛋白表达水平。采用RT-PCR方法检测各组孕妇胎盘组织中的αv和β3的mRNA表达水平。结果αvβ3免疫反应产物吸光度测定半定量分析结果:对照组(0.59±0.05),轻度PE组(0.38±0.04),重度PE组(0.22±0.03);Western blot检测各组胎盘组织αvβ3蛋白表达结果:对照组(1.39±0.12),轻度PE组(1.01±0.13),重度PE组(0.56±0.08);RT-PCR方法检测各组胎盘组织αv及β3mRNA水平结果:αvmRNA对照组(1.05±0.12),轻度PE组(0.99±0.10),重度PE组(0.97±0.10);β3mRNA对照组(0.86±0.09),轻度PE组(0.84±0.08),重度PE组(0.83±0.08)。3组均数间采用最小显著差数法(LSD法)进行两两比较,结果表明,与对照组相比,PE组孕妇胎盘组织中αvβ3蛋白表达低下,差异有统计学意义(P<0.05);其中重度PE组αvβ3蛋白表达水平更低,其与轻度PE组比较,差异有统计学意义(P<0.05)。Spearman相关性分析rs=-0.931,P<0.01,表明αvβ3蛋白与子痫前期病情的严重程度呈显著负相关,即随病情加重其表达进一步降低。但αv和β3的mRNA的表达水平3组间比较差异无统计学意义(P>0.05)。结论整合素αvβ3在PE胎盘组织中的低表达可能参与了子痫前期的发病过程,并能反映子痫前期病情的严重程度。
Objective To study the mRNA and protein expression of integrin αvβ3 in the placental tissues from healthy pregnant women and the patients with preeclampsia,and explore the relationship between αvβ3 and preeclampsia.Methods Immunohistochemistry,Western blot and RT-PCR were used to detect the expression of αvβ3 protein and mRNA in the placenta of 25 women with preeclampsia(including 10 cases of mild preeclampsia,and 15 cases of severe preeclampsia)and 20 normal full-term pregnant women,respectively.Results Semi-quantitative analysis showed that,the average absorbance(A)values of αvβ3-immunoreactive products in control group,mild PE group and severe PE group were(0.59±0.05),(0.38±0.04),(0.22±0.03),respectively.Western blot results revealed that A values of αvβ3 protein in control group,mild PE group and severe PE group were(1.39±0.12),(1.01±0.13) and(0.56±0.08),respectively.The A values of αv mRNA in placental tissues of control group,mild PE group and severe PE group were(1.05±0.12),(0.99±0.10),(0.97±0.10) respectively.The A values of β3 mRNA in placental tissues control group,mild PE group and severe PE group were(0.86±0.09),(0.84±0.08),(0.83±0.08) respectively.As compared with control group,the protein expression levels of integrin αvβ3 were significantly decreased in preeclampsia groups(P〈0.05),and there was significant difference between severe and mild PE groups(P〈0.05).Spearman correlation analysis showed the correlation coefficient was-0.931(rs=-0.931),P〈0.01,suggesting that the expression of αvβ3protein was significantly negatively related to the severity of PE.However,there was no significant difference in the expression of αv and β3 mRNA among three groups(P〈0.05).Conclusion The decreased expression of integrin αvβ3 may be involved in the pathogenesis of PE,and can reflect the severity of PE.
