摘要
目的:建立高效液相色谱测定乌药叶中槲皮素含量的方法。方法:色谱柱(ZORBAX ECLIPSE XDB-C18,150 mm×4.6 mm,5μm),以甲醇∶0.4%磷酸=50∶50为流动相,流速:1.0 mL/min,检测波长:360 nm,柱温:30℃。结果:槲皮素在0.129μg~1.29μg内线性关系良好(r2>0.9999);重复性RSD为4.2%;平均加样回收率为101.0%,RSD为2.37%。结论:该方法专属性强,简便快捷,结果准确,可用作乌药叶槲皮素的含量测定。
Objective To establish a method for the determination of quercetin in the leaves of Lindera aggregate(Sims) Kosterm.Methods Colum ZORBAX ECLIPSEXDB-C18,150 mm×4.6 mm,5 μm,the mobile phase consisted of methanol-0.4% phosphoric acid 50∶50 with a flow rate of 1.0 mL/min,the detection wavelength was monitored at 360 nm,and the column temperature was maintained at 30℃.Results The calibration curves of quercetin were linear in the range of 0.129 μg^1.29 μg.The reproducibity RSD was 4.18%,the average recovery of quercetin was 101.0%(RSD=2.37%).Conclsion The method is specific,simple,quick,accurate and can be used for the determination of quercetin in leaves of Lindera aggregata.
