摘要
目的探讨影响壳聚糖(CS)纳米粒包封卵蛋白(OVA)的因素以优化包封条件,初步研究OVA体外释放规律,观察CS缓释OVA的作用。方法采用离子凝胶法,以三聚磷酸钠(TPP)为交联剂,制备CS纳米粒并包封OVA,检测其表征参数。观察CS浓度、CS/OVA质量比、CS/TPP质量比和CS溶液pH值对CS纳米粒包封OVA的影响,以及载OVA的CS纳米粒的OVA体外释放。结果载OVA的CS纳米粒平均粒径为496.7nm,Zeta电位为+29.41mV。增加CS浓度、CS溶液pH值和CS/OVA质量比,降低CS/TPP质量比可以增加CS对OVA的包封率。载OVA的CS纳米粒呈缓释模式,体外释放可持续6天以上。结论 CS浓度、CS溶液pH值、CS/OVA质量比和CS/TPP质量比是影响CS纳米粒对OVA的包封率的重要因素。CS纳米粒具有缓释OVA作用。
Objective To investigate the factors which influence the encapsulation efficiency and examine in vitro the release of ovalbumin (OVA)-loaded ehitosan (CS) nanoparticles. Methods The CS nanopartieles were prepared by ionotropic gelation of cross-linking of CS with sodium tripolyphosphate (TPP) , and OVA was encapsulated by CS nanoparticles. The effects of CS concentration, CS/OVA mass ratio, CS/TPP mass ratio and initial pH value of CS solution on the encapsulation efficiency of OVA-loaded CS nanoparticles were evaluated, and in vitro release was detected. Results Average size of the OVA-loaded CS nanoparticle was 496.7nm, and Zeta potential was + 29.41inV. High CS concentration, high CS/OVA mass ratio, high pH value of CS solution and low CS/TPP mass ratio all leaded to high encapsulation efficiency of OVA-loaded CS nanoparticles. OVA-loaded CS nanoparticles revealed a controlled release of OVA and had a continuous release of 6 day in vitro. Conclusions CS concentration, CS/OVA mass ratio, CS/TPP mass ratio and initial pH value of CS solution are the important factors which influence the encapsulation efficiency of OVA-loaded CS nanoparticels. OVA loaded by CS nanoparticles releases slowly.
