摘要
目的探讨阴道超声照射对人胚绒毛滋养细胞Bcl-2及活化Caspase-3蛋白表达的影响,以及在滋养细胞凋亡中的作用。方法将符合条件的患者50例,随机分为5组,每组10例。4组实验组经阴道超声持续照射10 min后,分别于1 d、3 d、5 d、7 d人工流产,收集绒毛标本,第5组为对照组。采用免疫组织化学SABC法,检测各组绒毛滋养细胞中的Bcl-2蛋白及活化Caspase-3蛋白的表达情况。结果随取胚天数的后延,Bcl-2蛋白表达的量逐渐增加、活化Caspase-3蛋白表达的量逐渐减少;Bcl-2蛋白、活化Caspase-3蛋白表达的量分别于辐照后第3天、第5天始与对照组比较差异无统计学意义(P>0.05)。结论随着阴道辐照时间的推移,细胞凋亡减少,同时人胚绒毛滋养细胞的凋亡指数与其Bcl-2蛋白、活化Caspase-3蛋白的表达呈相关性。
Objective To investigate the expression of Bcl-2 protein and activated Caspase-3 protein of human chorionic villus cytotrophoblasts exposed to transvaginal ultrasound irradiation,and its role in the apoptosis of trophocyte.Methods Fifty matched patients were randomly divided into 5 groups.Four experimental groups were being respectively obtained the chorionic villus at one day,three days,five days,7 days after being exposed to transvaginal ultrasound irradiation for 10 minutes in the first trimester of pregnancy.The fifth was control group(bland radiation).The immunohistochemistry(SABC) was used to detect the expressions of Bcl-2 protein and activated Caspase-3 protein of human trophocyte in both groups.Results With the development of embryos,the expression of Bcl-2 protein ascended while the expression of Caspase-3 protein gradually declined.It was three days when the expression of Bcl-2 protein began to be no significant difference between experimental group and control group(P0.05) after being exposed.And it was five days when the expression of Caspase-3 protein did so.Conclusion With the increasing of irradiation time,the apoptotic index gradually declined.There was an association between the apoptotic index and the expressions of Bcl-2 protein and activated Caspase-3 protein in human trophocyte.
