冠心病患者缺氧诱导因子-1α与血红素氧化酶-1的相关性研究 被引量:9

摘要 目的探讨冠心病患者外周血单个核细胞缺氧诱导因子-1α(HIF-1α)与血红素氧化酶-1(HO-1)的相关性。方法 选择经冠脉造影确诊的冠心病患者为病例组,按照临床表现分为急性心肌梗死组(AMI组)、不稳定型心绞痛组(UAP组)和稳定型心绞痛组(SAP组);选择冠脉造影未发现冠脉病变的患者为对照组。利用Western blot检测入选对象外周血单个核细胞HIF-1α和HO-1的蛋白表达水平,相关分析方法分析两者表达水平的相关性。结果与对照组相比,AMI和UAP组HO-1和HIF-1α蛋白表达均明显增高,SAP组患者增高不明显。总体水平上,HIF-1α与HO-1呈正相关(r=0.76);在AMI、UAP、SAP组HIF-1α与HO-1表达水平也呈正相关。结论冠心病发生、发展过程中HIF-1α/HO-1通路被激活,HIF-1α可能通过诱导HO-1蛋白表达参与心肌保护作用。 Objective To investigate the correlation between hypoxia inducible factor-1α(HIF-1α) and heme oxygenase-1(HO-1) in patients with coronary artery disease. Methods A total of 95 patients with coronary artery disease(30 acute myocardial infarction,30 unstable angina pectoris,and 30 stable angina pectoris) and 30 control subjects with normal coronary artery were collected according to coronary angiogram.The protein expressions of HIF-1α and HO-1 in monocytes and lymphocytes were detected by Western blotting,and then the relationship between HIF-1α and HO-1 was investigated. Results Compared with the control group,the expression of HIF-1α and HO-1 in AMI and UAP group increased significantly,but increased slightly in SAP group.There was a positive correlation between the expression of HIF-1α and HO-1 in the total level(r=0.76) as well as in each case group. Conclusion The HIF-1α/HO-1 pathway is activated in the development of coronary artery disease,and HO-1 is induced by HIF-1 to participate in the cardioprotection.
