摘要
目的研究承德产穿山龙药材的HPLC指纹图谱并测定薯蓣皂苷元的含量。方法选择10批承德不同产地的穿山龙,用Agilent Eclipse C18色谱柱,以水和乙腈为流动相,梯度洗脱,检测波长203 nm,柱温30℃,流速1.0 mL·min-1,分析时间90 min。结果不同产地的穿山龙药材中薯蓣皂苷元的含量差别较小,指纹图谱共标示出13个共有峰。利用中药色谱指纹图谱相似度评价系统研究版(2004A)软件,以夹角余弦为测度,测得10批穿山龙药材的指纹图谱相似度为0.90~1.00,说明各批穿山龙药材之间既有较好的相关性,又有区别。结论 HPLC指纹图谱可整体显示穿山龙药材的特征,为穿山龙的质量控制提供了有效手段。
OBJECTIVE To establish an HPLC method for the determination of diosgenin,and establish a chromatographic fingerprint of Dioscorea nipponica Makino.METHODS 10 batches of Dioscorea nipponica Makino of Chengde from different habitats were chosed.An HPLC was performed on an Agilent Eclipse C18 column,the mobile phase used in gradient elution consisted of water and acetonitrile,with the flow rate of 1.0 mL·min-1.The column temperature was 30 ℃ and the wavelength was 203 nm.The analysis time was 90 min.RESULTS There was little difference in the content of diosgenin in Dioscorea nipponica Makino.13 common peaks on the HPLC fingerprints of them were marked.A software was uesd,it is a similarity evaluation system for chromatographic fingerprint of TCM(Version 2004A).The simlarities were determined by cosine.The results of similarity analysis were 0.90-1.00.It had good correlation with each batch and accompanied difference.CONCLUSION The characteristics of Dioscorea nipponica Makino of Chengde from different habitats were obtained on the whole and the method can be used for the quality control of Dioscorea nipponica Makino.
