摘要
目的比较丹参酮ⅡA与丹参酮ⅡA磺酸钠对人白血病K562细胞的体外增殖抑制与诱导分化作用。方法采用改良MTT法测定两种药物对K562细胞的增殖抑制作用,倒置显微镜下观察其对K562细胞形态的影响,以硝基四氮唑蓝还原试验法,用流式细胞仪测定细胞膜表面的分化抗原CD33和CD11b,观察丹参酮ⅡA与丹参酮ⅡA磺酸钠对K562细胞的诱导分化作用。结果 3.4~13.6μmol·L-1丹参酮ⅡA对K562细胞有显著的增殖抑制作用,且呈时间和剂量依赖性;丹参酮ⅡA对K562具有诱导分化作用,表现为NBT还原能力增强,CD33表达下降,CD11b表达升高。丹参酮ⅡA磺酸钠对K562细胞无增殖抑制与诱导分化的作用。结论丹参酮ⅡA对K562细胞具有增殖抑制与诱导分化作用,磺酰化后其药理活性丧失。
OBJECTIVE To investigate inhibiting effect and inducing differentiation of tanshinone ⅡA and sodium tanshinone ⅡA sulfonate on human erythroleukemia K562 cells in vitro.METHODS The inhibiting effect of tanshinone ⅡA and sodium tanshinone ⅡA sulfonate on the K562 cells was evaluated by modified MTT assay method,and the cell morphology was observed by inverted phase contrast microscope.The cell differentiation was evaluated by NBT reduction assay and the measurement of CD33 and CD11b was performed by flow cytometric analysis.RESULTS In tanshinone ⅡA groups(3.4-13.6 μmol·L-1),the proliferation of K562 cells was significantly suppressed in a time-and dose-dependent manner,and K562 cells were differentiated into mature,which was exhibited as the increase of the NBT reducing capacity and the expression of CD11b,as well as the decrease of the expression of CD33.However,neither growth inhibition nor differentiation induction was observed on K562 cells in sodium tanshinone ⅡA sulfonate group.CONCLUSION Tanshinone ⅡA shows a marked proliferation inhibiting effect and inducing differentiation on K562 cells but sodium tanshinone ⅡA sulfonate has no anti-tumor effect to K562 cells.
