摘要
目的应用纤维环穿刺法与腰椎失稳法建立大鼠腰椎间盘退变模型,比较两种方法的异同。方法针刺组选用21G穿刺针对大鼠L3,4、L4,5、L5,6椎间盘行全层针刺;失稳组切除大鼠L3~L6棘突、棘上、棘间韧带和两侧关节突。于术后2周、4周和8周每组各取6只大鼠行腰椎影像学和组织学检查。结果 针刺组大鼠2周时出现椎间隙狭窄,纤维环断裂,T2信号强度减低;4周时退变进展;8周时出现髓核皱缩、纤维化,T2呈完全低信号。失稳组4周时出现退变征象,8周时退变加重。结论 两种方法均能建立大鼠腰椎间盘退变模型,针刺法较失稳法退变出现早、进展快且程度严重。
Objective To establish two kinds of rat intervertebral disc degeneration models by anulus puncture and lumbar instabili- ty,and to compare their respectively of characteristics. Methods In the puncture group,intervertebral disc of L3,4,L4,5 and L5,6 were exposed and using a 21-gauge needle to make a full-thickness puncture. In the lumbar instability group,resected rats,spinous process, interspinous ligament,supraspinous ligament and zygapophysis of L3- L6 vertebra. Taked 6 SD rats each group to examine their lumbar intervertebral disc at 2,4,and 8 weeks after operation with radiological and histopathology methods. Results In the puncture group observed following signs:the height of intervertebral space narrowing,annulus rupture,T2-weightcd image declined at week 2;the degen- eration signs was increased at week 4;nucleus appeared shrinkage and fibrosis,T2-weighted image presented completely low signal at week 8. In the lumbar instability group,signs of degeneration appeared at week 4;degeneration was progressed at week 8. Conclusion The rat model of intervertebral disc degeneration can be successfully constructed by the methods of anulus puncture and lumbar instability. The degenerative process induced by anulus puncture is faster and more severe than the induced by lumbar instability.
