摘要 目的探讨流式细胞仪每秒进样细胞数对被检测细胞的细胞周期影响。方法以鸡肝细胞为实验对象,采用不同的每秒进样细胞数检定其DNA含量,以MultiCycle for Windows分析软件分析G0/G1期、S期、G2/M期比值的变化。结果 G0/G1期细胞值方差分析显示,F=2.7,P〉0.05;S期细胞值方差分析,F=11.6,P〈0.05,G2/M期细胞值方差分析,F=17.4,P〈0.05。表明每秒进样细胞数不同,其细胞的S期、G2/M期比值之间的差异有显著性。结论每秒进样细胞数影响细胞的S期、G2/M期比值,每秒进样细胞数以150~400个为宜。 Objective To investigate the effect of the cell number of sample injection per second on cell cycle.Methods Fowl liver cells were prepared and their DNA content was assayed by different cell numbers of sample injection per second.MultiCycle for Windows software was used for analyzing cell rates of G0/G1,S and G2/M phrase.Results The statistical analysis indicated that the cell rates of both S and G2/M phrase had significant differences,but G0/G1 phrase cell rate had no statistical significance(F=11.6,P0.05;F=17.4,P0.05;F=2.7,P0.05) among these groups' different cell numbers of sample injection per second.Conclusion The cell number of sample injection per second affects cell rates of S and G2/M phrase and the range from 150 to 400 cells per second was suitable for cell cycle analysis.
