摘要
目的研究大鼠牙髓干细胞的细胞周期及其核型特征。方法采用流式细胞仪对处于对数生长期的大鼠牙髓干细胞进行细胞周期的检测,并采用ModFIT LT周期分析软件对其进行分析。采用胰蛋白酶-Giemsa染色法对大鼠牙髓干细胞进行核型染色并进行统计分析。结果大鼠牙髓干细胞的G1期细胞比例为75.24%,S期为23.20%,而大鼠骨髓间充质干细胞G1期比例为66.80%,S期为28.79%,两种细胞的G2/G1比值均为2.0;核型分析大鼠牙髓干细胞正常二倍体比例为91%。结论与大鼠骨髓间充质干细胞相比,牙髓干细胞的增殖能力较弱。两种细胞的分裂方式基本以二倍体方式进行。核型分析结果表明,所获得的2B2株大鼠牙髓干细胞核型稳定。
Objective To study the characteristics of cell cycle and karyotype of 2B2 rat dental pulp stem cells. Methods The cell cycle of 2B2 cells was assayed by FCS and analyzed using ModFITLT software. Trypsin- Giemsa staining was performed for karyotype analysis. Results 2B2 rat dental pulp stem cells at the stages of G1 and S accounted for 75.24 % and 23.20%, respectively, while bone marrow mesenchymal stem cells at the stages of G1 and S 66.80% and 28.79%, respectively. The G2/G1 was 2.0 for both of the two cells. Karyotype analysis indicated that diploid cells of 2B2 cells constituted 91%. Conclusions The capacity for proliferation of 2B2 rat dental pulp stem cells is less potential than bone marrow mesenchymal stem cells. The cells have a stable karyotype and divide in the manner of diploid.
