摘要
目的通过检测阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者血清谷胱甘肽硫转移酶(GSH-ST)活力、活性氧(ROS)水平的变化,探讨OSAHS患者体内氧化应激状态和抗氧化酶的变化及其临床意义。方法选择OSAHS患者(OSAHS组)及健康人(对照组)各30例,两组受检者的性别、年龄和体质指数间有均衡性。所有受检者在夜间入睡前及晨醒后抽取静脉血,用比色法测定各组GSH-ST活力及抑制羟自由基(OH.)能力、抗超氧阴离子自由基(O2-)能力。结果 OSAHS组睡前血清抑制OH.能力、抗O2-能力较对照组降低,OSAHS组这2个指标晨起比睡前降低,差异均有统计学意义(P<0.01);OSAHS组血清GSH-ST活力较对照组增高(P<0.01),OS-AHS组血清GSH-ST活力晨起与睡前无明显变化(P>0.05);OSAHS组睡眠呼吸暂停低通气指数(AHI)与晨起血清抑制OH.能力、抗O2-能力均呈负相关,与血清GSH-ST活力均呈正相关;OSAHS组血清GSH-ST活力与晨起抑制OH.能力、抗O2-能力呈负相关。结论 OSAHS患者有持续的氧化应激存在,并与OSAHS严重程度相关;OSAHS患者GSH-ST活力增高,且与OSAHS严重程度及氧化应激的程度有相关性。
Objective To evaluate the clinical significance of the change of oxidative stress status and antioxidase in patients with OSAHS through determination of the serum levels of reactive oxygen species(ROS) and glutathione S-transferases(GSH-ST) activity in patients with obstructive sleep apnea-hypopnea syndrome(OSAHS).Methods Patients with OSAHS diagnosed by Polysomnography(PSG) and healthy objects excluded of OSAHS by history and portable PSG,30 each,were enrolled.There were no significant differences in terms of age,sex and BMI between the two groups.Venous blood samples were obtained from all subjects before and after sleep-breathing monitoring.Activity of GSH-ST,the ability to inhibit hydroxyl radicals and superoxide anion radicals were measured by colorimetric technique.Results The serum abilities of inhibitiing hydroxyl radicals and superoxide anion radicals before sleep were lower in patients with OSAHS than in the control group,whereas within the OSAHS group itself,the after sleep levels were lower than those before sleep(P0.01).Activity of GSH-ST were higher in patients with OSAHS compared with those in control group(P0.01),and there were no differences between serum GSH-ST activity before and after sleep within the OSAHS group(P0.05).In the OSAHS group,AHI were negatively correlated to the serum ability of inhibiting hydroxyl radicals and superoxide anion radicals after sleep(r=-0.548,P0.01;r=-0.385,P0.05)and positively correlated to GSH-ST activity(r=0.789,P0.01).Serum GSH-ST activity were correlated negatively to the serum ability of inhibiting hydroxyl radical and superoxide anion after sleep(r=-0.509,P0.01;r=-0.610,P0.01).Conclusion OSAHS patients exhibit a sustained oxidative stress which is related to the severity of the condition,as well as a increased GSH-ST activity in serum which is correlated to both severity of the disease and extent of the oxidative stress.
