摘要
基于不同质量浓度Na2CO3溶液、反应温度和时间对反应体系吸光度影响单因素试验基础上,运用二次回归正交旋转组合设计方法优化反应条件;以微量移液器为工具,建立一种简化、微量测定葡萄与葡萄酒总酚含量的方法。结果表明,建立的回归模型达显著水平,利用优化函数途径进行模拟选优得到最佳反应条件为:0.5 mL样品,0.5mL 1 mol·L-1 Folin-Ciocalteu试剂(F-C试剂)和4.0 mL 52 g·L-1 Na2CO3溶液,在25℃下置102 min后,测765 nm吸光度。相对没食子酸质量浓度在0~0.050 g·L-1内,吸光度和没食子酸浓度有良好的线性关系(R2=0.999 4)。对酿酒葡萄赤霞珠与王朝干红葡萄酒样的加样回收率分别是98.4%和97.8%(n=3),相对标准差(RSD)分别为1.32%和2.01%。该法是一种简化、微量经济测定葡萄与葡萄酒总酚含量的方法,且具有较高的稳定性和准确度。
A simple method to determine total phenolic compounds in grape and wine was established.The reaction conditions were optimized through quadratic regression rotational combination design basing on the single factor experiments such as Na2CO3 concentration,reaction temperature and reaction time using pipette.The orthogonal experimental results showed that the optimum reaction system including 0.5 mL sample,0.5 mL 1 mol·L-1 Folin-Ciocalteu reagent and 4.0 mL 52 g·L-1 Na2CO3 incubated at 25 ℃ for 102 min,then determined the extinction value at 765 nm.The calibration curve showed a good linearity between the absorbance value and gallic acid concentration range from 0 g·L-1 to 0.050 g·L-1(R2=0.999 4).The average recovery rate evaluated by testing the berry of Cabernet Sauvignon and wine at three concentration(low,middle and high) were 98.4% and 97.8%(n=3),respectively,with the corresponding relative standard deviation(RSD) of 1.32% and 2.01%.It's a simple and economic method for phenolic compound determination in the grape and wine with high stability and accuracy.
