六溴环十二烷(hexabromocyclododecane,HBCD)暴露对生长阶段文昌鱼的毒性及其几种重要酶活性的影响 被引量:12

摘要 以文昌鱼为研究对象,研究了在六溴环十二烷暴露下对文昌鱼造成的机体氧化损伤.通过检测相关酶活变化情况,如酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD),从而确定低剂量长期暴露HBCD对文昌鱼鱼体产生氧化损伤效应的机制.急性毒性实验结果表明,文昌鱼暴露于六溴环十二烷饱和溶液(8.6ng·mL-1)96h后未有死亡发生,说明该物质毒性相对较小;亚慢性毒性试验表明,梯度暴露实验设置浓度分别为1.00、2.15、4.30、8.60ng·mL-1,暴露时间为14d,按体长将处于Ⅱ龄(2.0~2.8cm)和Ⅲ龄(3.2~4.0cm)的文昌鱼分别投毒检测,实验结果与对照组比较:Ⅱ龄文昌鱼的ACP、ALP、SOD活性均在1ng·mL-1浓度范围内受促进作用显著,2.15ng·mL-1时受抑制作用显著,然后随浓度升高酶活升高.CAT在2.15ng·mL-1时受促进作用显著,当浓度大于2.15ng·mL-1时受抑制作用显著,但并无明显剂量关系.Ⅲ龄文昌鱼总体上酶活变化较小,也无剂量关系.结果表明HBCD对Ⅱ龄文昌鱼的4种酶活影响比Ⅲ龄显著,与对照组相比,所有暴露组文昌鱼体重增长缓慢(p<0.05). The objective of this study was to evaluate the sub-lethal toxicity of the fire retardant hexabromocyclododecane(HBCD) in fish.Amphioxuses,as in vivo models,were exposed to waterborne HBCD from 1 to 8.6 ng · mL-1 for 14 days.Acid phosphatase(ACP),alkaline phosphatase(ALP),catalase(CAT),and superoxide dismutase(SOD) activities were measured.The results of acute toxicity experiments show that there were no amphioxus deaths after 96 h exposure in saturated solution of HBCD(8.6 ng · mL-1).The gradient exposure concentrations of the subchronic toxicity experiment were set at 1.00,2.15,4.30,8.60 ng · mL-1,and exposure time was 14 days.Age Ⅱ(2.0~2.8 cm) and age Ⅲ(3.2~4.0 cm) amphioxus were poisoned and detected separately.Compared to the control group,ACP,ALP,and SOD of age Ⅱ are promoted significantly at a concentration of 1 ng · mL-1,and inhibited significantly at 2.15 ng · mL-1;while in the case of CAT,significant promotion is detected at a concentration of 2.15 ng · mL-1.When the concentration is higher than 2.15 ng · mL-1,significant inhibition is observed.However,no visible relationship was found between dose and response.To the age Ⅲ,there are no significant changes for all the enzymes,which indicates that the effect of HBCD on age Ⅱ is significantly higher than on age Ⅲ.In addition,body weight increases more slowly compared to the control group(p0.05).
