大粒油莎豆块茎中淀粉和蛋白质提取工艺研究 被引量:6

摘要 以碘显色法测定了油莎豆块茎淀粉含量,以考马斯亮蓝G-250法测定蛋白质含量;利用石灰水浸泡法同时从油莎豆中提取出了淀粉和蛋白质,研究料液比、浸泡液pH值、浸泡时间、上清液pH值等因素对大粒油莎豆淀粉、蛋白质提取率的影响,并通过正交实验确定最佳提取工艺参数。结果表明:大粒油莎豆中淀粉含量为31.8%,蛋白质含量为2.9%;提取淀粉的最佳工艺条件为:浸泡液pH 10.0,料液比1∶5(W∶V),浸泡时间90 min;提取蛋白质的最佳工艺条件为:浸泡液pH 10.5,料液比1∶5,浸泡时间60 min,上清液在pH 5.0条件下沉淀蛋白。 The content of starch and protein in tubers of Cyperus esculentus was determined by the iodine-vapor staining method and coomassie brilliant blue staining method,respectively.Results showed that the content of starch was 31.8%,that of protein was 2.9%.Using the limewater extraction method,the starch and protein from chufa tubers were extracted.Through the individual factor test and orthogonal design test,some factors,such as the limewater pH value,the ratio of sample to solution(W:V),the immersion time,the supernate pH value,etc.were investigated.The optimal conditions for starch extraction were as follows: limewater pH 10,the ratio of sample to solution1:5,the immersion time 90 min.The optimal conditions for protein extraction were as follows: limewater pH 10.5,the ratio of sample to solution 1:5,the immersion time 60 min,protein deposition at the supernate pH 5.0.
