摘要
目的通过DEK基因沉默对CaSki细胞生长、衰老的影响,探讨DEK原癌基因在宫颈癌发生中的作用。方法将DEKsiRNA真核表达载体转入CaSki细胞,观察转染后48 h细胞形态学变化,测定细胞生长曲线、SA-β-半乳糖苷酶细胞化学染色鉴定细胞衰老。结果与未转染和转染psiRNA-hH1neo组相比,转染psiRNA-hHDEK组CaSki细胞的生长速度明显降低;psiRNA-hHDEK转染组SA-β-半乳糖苷酶染色阳性细胞百分率(83.2±9.46)%高于转染psiRNA-hH1neo组(18.9±6.46)%和未转染CaSki组(23.1±7.28)%,结果具有统计学意义(P<0.05)。结论 DEK基因沉默后CaSki细胞发生衰老,证明DEK基因在宫颈癌中是衰老抑制基因。
Objective To study the effect of DEK gene in cervical cancer CaSki cell by observing of growth and senescence in CaSki cells after DEK gene silencing.Methods Cell morphology was observed under light microscope at 48 h post DEK siRNA transfection,at the same time cell growth curve was tested and SA-β-galactosidase enzyme cytochemistry identified of cell senescence.Results The growth rate of CaSki cells of psiRNA-hHDEK transfection decrease significantly by comparing CaSki cells of non transfected and psiRNA-hH1neo ransfected;SA-β-galactosidase positive staining percentage of psiRNA-hHDEK transfected was(83.2±9.46)%,higher than psiRNA-hH1neo transfection group and the non-transfected,there was statistical significance(P0.05).Conclution DEK gene silencing can induce CaSki cells into senescence,which is an aging suppressor gene in cervical cancer.
