摘要
对透明质酸含量检测方法CTAB比浊法和Bitter-Muir法进行了优化,并用于发酵液中透明质酸含量的检测。优化结果为:CTAB比浊法最佳检测波长为400 nm,CTAB溶液质量浓度为5 g/L,反应温度为室温,络合时间为3 min,回归线性方程y=0.007 1x-0.101 8,R=0.999 9,发酵液中加标平均回收率101.29%,平均RSD值为0.70%;Bitter-Muir法最佳检测波长为540 nm,水浴时间为10 min,线性回归方程y=0.005 8-0.199 3x,R=0.996 5,发酵液中加标平均回收率83.87%,平均RSD值为2.70%。结果表明,用于发酵液中透明质酸含量的检测,CTAB比浊法更准确、更精确、更高效。
The Bitter-Muir method and CTAB turbidimetric method were optimized to assay the concentration of hyaluronic acid in fermentation broth. The optimum conditions of CTAB method are wavelength 400 nm,5 g/L CTAB,room temperature,and reaction time 3 min.The regression equation is y=0.007 1x-0.101 8,R=0.999 9.The precision and recovery of this method are within 0.70% and 101.29%,respectively.The optimum conditions of Bitter-Muir method are wavelength 540 nm,reaction time 10 min.The regression equation is y=0.005 8-0.199 3x,R=0.996 5.The precision and recovery of this method are within 2.70% and 83.87%,respectively.The results showed that CTAB method is superior to Bitter-Muir method in terms of accuracy,precision and efficiency.
