摘要
目的:利用胶滴药敏试验评估人胃中分化腺癌细胞株SGC-7901、人胃低分化腺癌细胞株BGC-823和人胃黏液腺癌细胞株MGC-803对化疗药物的敏感性。方法:利用3个胃癌细胞株建立胶滴药敏试验,每个细胞株用顺铂、奥沙利铂、5-氟尿嘧啶(FU)、依托泊苷、阿霉素、丝裂霉素6种化疗药物测定胃癌细胞的敏感性和抑制率。结果:6种化疗药在标准浓度即100%测试药物浓度(TDC)下对人胃黏液腺癌细胞株MGC-803的抑制率分别为58.68%、63.85%、57.60%、28.09%、23.18%和40.18%,对人胃低分化腺癌细胞株BGC-823的抑制率分别为59.09%、61.26%、56.30%、33.21%、33.26%和41.20%,对胃中分化腺癌细胞株SGC-7901的抑制率分别为61.25%、69.76%、60.78%、35.00%、30.08%和58.80%。结论:体外胶滴药敏试验是一种较好的药敏检测方法,对化疗药物的体外筛选和个体化治疗具有实际应用价值。
Objective: To evaluate the sensitivity of three different gastric cancer cell lines to the chemotherapy drugs with the use of the collagen gel droplet embedded-culture drug sensitivity test (CD-DST). Methods: Three gastric cancer cell lines of SGC-7901, BGC-823 and MGC-803 were utilized to establish CD-DST. Six chemotherapy drugs, including cisplatin, oxaliplatin, 5-fluorouracil (FU), etoposide, adriacin and mitomycin were used to measure the sensitivity and inhibition ratio in each of cancer cell lines. Results: Under standard concentration (TDC, 100% test drug concentration), the inhibition ratios of six chemotherapy drugs to the human gastric mucous adenocarcinoma cell line MGC-803 were 58.68%, 63.85%, 57.60%, 28.09%, 23.18% and 40.18%, to the poorly differentiated human gastric adenocarcinoma cell line BGC-823 were 59.09%, 61.26%, 56.30%, 33.21%, 33.26% and 41.20%, and to moderately differentiated adenocarcinoma cell line SGC-7901 were 61.25%, 69.76%, 60.78%, 35.00%, 30.08% and 58.80%. Conclusion:CD-DST is a better method for testing drugsensitivity , which is of practical value to extracorporeal anti-cancer drug screening and individualized treatment.
