导数同步荧光法测定血清色氨酸 被引量:3

Determination of tryptophan in serum by derivative synchronous fluorescence method

摘要 为了排除酪氨酸、苯丙氨酸对荧光法测定色氨酸的干扰,建立了色氨酸导数-同步荧光测定法。结果表明,当Δλ为90nm时,色氨酸的一阶导数-同步荧光峰在285nm时不受酪氨酸、苯丙氨酸的干扰;色氨酸浓度在0~400μmol/L与导数-同步荧光值成线性,Y=0.287X±0.096,r=0.999;批内、批间CV分别为2.57%、3.42%;回收率93.8%~97.2%;血红蛋白含量在0.25~5.0g/L、胆红素含量在42.75~342μmol/L对测定无干扰;30例健康成年人血清色氨酸测定结果为73.98±7.26(x±s)μmol/L;本法操作简单、快速,测定结果准确,是血清色氨酸常规测定较为理想的方法。 To remove the interference from tyrosine and phenylalanine in the determination of tryptophan by fluorescence method,The author established the determination method of free tryptophan in serum by derivative synchronous fluorescence.The results showed,when was 90 nm,the peak of derivative synchronous fluorescence wasn't disturbed by tyrosine and phenylalanine at 285 nm. The linear rang is 0 to 400 mol/L(Y=0287X0096,r=0999).The intraand interassay CV are 257% and 342%,respectively.The analytical recovery is 938% to 972%. The determination wasn't disturbed by hemoglobin(02550 g/L)and bilirubin(4275342 mol/L).Reference value of the tryptophan in serum from 30 healthy individuals were 7398726( s ). The method is simple,rapid,and precise,so it is a reasonable method for detection of tryptophan in serum.
