摘要
目的 观察氯化锂协同温热对 B G C823 人胃癌细胞的生物学效应。方法 通过克隆形成率、细胞生长曲线测定肿瘤细胞生长特性变化,用免疫组化染色测定细胞 Hsp70、p53 表达变化。结果 研究表明氯化锂在一定范围内随着浓度的增加,对肿瘤细胞生长的抑制作用逐渐增强;41℃温热对肿瘤细胞无显著抑制作用;两者合并作用时,克隆形成率进一步降低( P< 0.01),显示明显协同效应。免疫组化观察显示, Hsp70 主要分布于细胞质,41℃温热可诱导 B G C823 细胞 Hsp70 表达增加,但温热与氯化锂合并作用则使 Hsp70 的表达显著抑制( P< 0.05);p53主要分布于细胞核,氯化锂(1 m g/m l)及合并作用对p53 表达有轻度抑制作用。结论 单价阳离子锂与温热合并有一定协同抑制肿瘤细胞生长作用。其中抑制 Hsp70 表达,阻止热耐受的产生可能是其协同机理之一。
Objective To observe the biological effects of lithium chloride in combination with hyperthermia on BGC 823 gastric cancer cell line.Methods Analyse the growth characteristics of the cell line by measuring the cloning efficiency and the cell growth curves;Detect the changes of expression of Hsp70 and p53 proteins by immunocytochemicstry.Results the inhibitory effects of lithium chloride on BGC 823 cell were gradually enhanced with the increasing of concentration of Li +.Lithium chloride combined with hyperthermia showed significantly synergetic effects,which made the cloning efficiency decrease further( P < 0.01 ).Immunocytochemical study showed that Hsp70 expressed in cytoplasm of BGC 823 cell.The expression of Hsp70 could be enhanced by hyperthermia at 41℃,but inhibited by hyperthermia in combination with lithium chloride( P <0.05);p53 protein expressed in cell nucleus,specially nuclear membrane,which was inhibited slightly by lithium chloride and the combination of hyperthermia and lithium chloride( P <0.05).Conclusions The results suggest that nuivalent cation combined with hyperthermia has the synergetic inhibitory effects on the growth of the tumor cells.The inhibition of Hsp70 expression,which might block thermotolerence,could be one of the mechanisms,for synergetic effect.\;
