姬松茸多糖对HT-29细胞与内皮细胞侵袭、黏附的影响和作用 被引量:5

Effect of Agarieus blazei Polysacharide on Invasion and Adhesion of Colon Carcinoma HT-29 to Endothelium

摘要 目的:探讨低分子量姬松茸多糖(LMPAB)对肿瘤细胞与内皮细胞侵袭和黏附的影响和作用。方法:不同剂量的LMPAB(5,10,20 mg.L-1)分别作用于人结肠癌细胞(HT-29)和人脐静脉内皮细胞(HUVECs)48 h,采用MTT法检测LMPAB对HT-29和HUVECs增殖的影响、采用分光光度法检测LMPAB对HT-29细胞与HUVECs黏附的影响、采用Transwell双层小室观察LPMPAB对HT-29迁移穿过单层HUVECs的影响。结果:不同剂量LMPAB(5,10,20 mg.L-1)对HT-29和HUVECs增殖无显著性作用;使HT-29黏附于单层HUVECs的数量得到了明显的抑制,抑制率分别为26.96%,23.60%,27.66%;对HT-29迁移穿过HUVECs也具有抑制作用,抑制率分别为23.96%,23.59%,41.36%。结论:低分子量姬松茸多糖通过抑制HT-29肿瘤细胞与内皮细胞的黏附以及肿瘤细胞迁移穿过内皮细胞而发挥其抗肿瘤转移作用。 Objective:To investigate the effects of low molecular weight polysaccharide lsolated from Agaricus blazei(LMPAB) on tumor cell adhesion and transmigration to HUVECs.Method: HT-29 cell and HUVECs proliferation were analyzed by MTT.Adhesion and transmigration ability of HT-29 to monolayer HUVECs were examined by spectrophotometry and Transwell chambers.Result: LMPAB(5,10,20 mg.L-1) showed no cytotoxic affect to HT-29 cells as well as the proliferation of HUVEC.LMPAB could significantly decrease counts of HT-29 adhesion to and transmigration through HUVECs.Conclusion: LMPAB exerts the anti-tumor metastasis activity via inhibition adhesion and transmigration of tumor cells toand through HUVECs.
