摘要
采用融粘帚霉AS3.3987对葛根素进行微生物转化,生成的主要产物经分析鉴定是3′-羟基葛根素。采用高效液相色谱方法,以XDB-C18为色谱柱,甲醇:水(27:73)为流动相,检测波长为254 nm,测得在160 r/min、30℃的转化条件下,底物加量为400μg/mL,转化时间为36 h时,3'-羟基葛根素得率最高为93.2 mg/g葛根素。
The major product of microbial transformation of puerarin by Gliocladium deliguescens AS3. 3987 was identi- fied as 3'-hydroxypuerarin on the basis of its spectral data. 3'-hydroxypuerarin was directly determined by HPLC. Methanol and water (27:73) was used as the mobile phase on C18 column and the UV detection wavelength was 254 nm. The maximum transformation rate obtained was 93.2 mg 3'-hydroxypuerarin/g puerarin at 30℃ 160 r/min for 36 h with the substrate concentration of 400μg/mL.
