摘要
目的观察大鼠失神经支配骨骼肌组织形态学及运动终板退变,寻找反映肌肉萎缩形态学指标。方法选用42只SD大鼠,切断胫神经建立失神经支配腓肠肌实验模型。运用电子天平测定肌肉湿重,图像分析仪测定肌细胞直径及截面积,氯化金压染法及透射电镜观察失神经支配骨骼肌超微结构及运动终板变化。结果萎缩肌肉湿重4周内下降最快,以后逐渐减慢并维持于一定水平,肌细胞直径及截面积呈持续性下降;运动终板4周内改变不明显,6周后退变逐渐加重,16周后消失,超微结构变化与光镜观察结果基本一致。结论肌肉湿重、肌细胞直径及截面积可作为反映肌肉萎缩形态学指标,后两者更为可靠。周围神经损伤后修复手术越早越好,力争在运动终板消失前进行。
Purpose To observe the degenerations of histo
morphology and motor end plate on rat denervated skeletal muscle,and look for the
morphological index evaluating the degree of muscle atrophy. Methods 42 SD rats were used
and the models of denervated skeletal muscle were established by cutting off the tibial
nerve.The muscle wet weight was weighed by electron balance,the diameter and cross section
area of muscle cell were measured with VIDⅢ image analysis system,and the changes of
motor end plate were observed with auric chloride pressing stain and transmission electron
microscope. Results The muscle wet weight most rapidly reduced within 4 weeks.It remained
about 30 per cent of normal value afterwards.The diameter and cross section area of muscle
cell reduced progressively along with the duration of denervation,especially within 4 weeks.The
motor end plate had not much change within 4 weeks.But the degeneration accelerated in 6
weeks and the end plate was not stained out in 16 weeks.The change of ultrastructure was
accord with the observation under light microscope. Conclusions The muscle wet weight,the
diameter and cross section area were considered evaluating index of muscle atrophy
degree.The former was more reliable than the latter.The repair operation of peripheral nerve
injury had better peform as soon as possible before the disappearance of motor end plate.
