摘要
目的检测不同病期强直性眷柱炎(ankylosingsporidylitis,AS)患者血清中Dickkopf-1(DKK—1)和肿瘤坏死因子(INF—a)的表达水平,初步探讨成骨相关因子表达水平与As的关系。方法采用酶联免疫吸附法(EUsA)检测47例As患者(包含22例早期患者和25例中晚期患者)、20例类风温性关节炎(RA)患者和20例健康对照人群血清DKK—1和TNF—a的表达水平。结果AS和RA患者血清中DKK—1和TNF—a水平明显高于正常对照,AS和RA患者组相比较血清DKK-1和TNF—a水平没有显著差异。中晚期As患者DICK—1水平较早期患者明显降低,INF—a表达也有降低,但无显著差异,Spearman分析显示,AS患者血清DKK-1和TNF—a表达水平正相关。结论随着AS骨化程度的进展,DKK—1表达呈下调趋势,提示DKK-1的表这下调可能参与了AS患者的骨化,DKK—1可能是与As骨化程度相关的血清标志物。
Objective To detect the expression of Dickkopf- 1 (DKK- 1) and tumor necrosis factor (TNF- a) in serum of ankylosing spondylitis with different stages in order to study the relationship of bone - related factors and AS. Methods The expression of DKK - 1 and TNF- a was detected by enzyme - linked immunosorbent assay (ELISA) in 47 eases of AS patients (22 cases of early stage and 25 cases of late stage), 20 eases of rheumatoid arthritis (RA) patients and 20 eases of healthy eontrol. Results The expression of DKK - 1 and TNF - a in AS and RA were significantly higher than control, but AS and RA were not significantly different. The expression of DKK - 1 in late stage was significantly decreased with early stage, TNF - a also decreased in late stage patients, but not significant. Spearman analysis suggests that DKK - 1 have positive correlative with TNF - a in AS. Conclusion With the progress of ossifieation in AS, the expression of DKK - 1 in a downregulated state, suggesting that the level of DKK - 1 may be involved in ossification of AS, DKK - 1 may be a potential marker which is associated with the degree of ossification in serum of AS.
