摘要
目的探讨大鼠肺缺血再灌注损伤时CXCL9因子的表达和中介素1-53(IMD1-53)对其的影响。方法建立大鼠在体肺缺血再灌注模型。将72只健康的Wistar大鼠随机分为3组:假手术组(C组)、缺血再灌注组(IR组)、中介素1-53组(IMD组)。每组取缺血45min、再灌注60min、再灌注120min时点各8只大鼠留取血浆及右肺组织。ELISA法检测血浆中CXCL9的含量,组织学评价肺组织损伤。结果 IMD组血浆中CXCL9因子的表达均低于IR组,组织学变化显示预先给予IMD1-53可减轻肺缺血再灌注损伤。结论 IMD1-53对肺缺血再灌注损伤有保护作用,其作用机制可能为减少趋化因子CXCL9因子的表达。
Objective To investigate the expression of CXCL9 in lung ischemia reperfusion injury in rats and the effect of IMD1-53 on it. Methods Rat lung model of ischemia reperfusion injury was established in vivo. Seventy-two healthy wistar rats were randomly divided into three groups: sham surgical control (C group), ischemia repelfusion group (IR group), IMD1-53 group (IMD group). The plasma sample and the right lung were collected at different three time points: 45 min ischemia, 60 min and 120 min reperfusion(8 rats in each group). The plasma content of CXCL9 was determined by ELISA and the lung tissue injury was evaluated by histology. Results The expression of CXCL9 in IMD group was significantly lower than other groups. Pretreated by IMD1-53, the lung tissue injury was descended through histology change. Conclusion IMD1-53 treatment before ischemia can protect the lung tissue against ischemia reperfusion injury possibly by reducing CXCL9 expression.
