摘要
目的:观察血小板衍生生长因子-BB(PDGF-BB)与胰岛素样生长因子-Ⅰ(IGF-Ⅰ)联合应用对大鼠正畸牙移动的影响,为正畸临床治疗加快正畸牙移动,缩短正畸治疗时间进行探索。方法:在32只SD雄性大鼠上颌安装施加50 g力正畸装置牵引上颌第一磨牙近中移动,隔日在正畸牙颊侧牙龈黏膜下分别或联合注射200 ng rhIGF-Ⅰ及10 ng rhPDGF-BB,对照组注射1%PBS。加力10 d后记录大鼠第一磨牙移动距离,处死动物,并取材用HE染色法观察牙周组织变化情况,抗酒石酸酸性磷酸酶染色(TRAP)计数压力侧破骨细胞数量。结果:PDGF-BB及IGF-Ⅰ均明显促进压力侧破骨细胞增殖,加速正畸牙移动,与对照组比较差异有统计学意义(P<0.01);PDGF-BB与IGF-Ⅰ联合应用有协同效应,其压力侧破骨细胞计数与对照组(P<0.01)及单一因子组(P<0.05)相比有明显差异,正畸牙移动距离与对照组(P<0.01)、IGF组(P<0.01)及PDGF组(P<0.05)比较差异有显著统计学意义。结论:外源性PDGF-BB和IGF-Ⅰ在正畸牙的局部联合应用较单独应用进一步促进牙周组织改建,加速正畸牙移动,具有协同效应。
Objective: To explore the combined effects of local injection of platelet-derived growth factor-BB(PDGF-BB)and insulin-like growth factor-I(IGF-I)in the periodontal tissues of rats during orthodontic tooth movement.Methods: The first upper left maxillary molar moved mesially by a closed-coil spring delivered a force of 50g in 32 male Sprague-Dawley rats,which were injected with 10ng PDGF-BB,200ng IGF-I alone or in combination in the buccal submucosal area of the molar.The injection was applied every other day.The distance of the tooth movement was measured after the experiment lasted for ten days,and then these rats were sacrificed.Sections were HE-stained for observing the activities of periodontal tissues rebuilding and TRAP-stained for counting the number of osteoclasts.Results: IGF and PDGF group all showed an increasing number of osteoclasts in the compressing side and orthodontic tooth movement rate compared with the control group(P0.01).The effect of combination of PDGF-BB with IGF-I was the most significant one,in which the number of osteoclasts in the compressing side and the tooth movement rate showed a significant increase compared with the control(P0.01),PDGF group(P0.05) and IGF group(P0.05,P0.01).Conclusion: The combination of exogenous PDGF-BB and IGF-I in orthodontic tooth movement may enhance the periodontal tissue remodeling and the orthodontic tooth movement rate compared with the application of two growth factors alone.So,PDGF-BB and IGF-I may have a synergistic effect during orthodontic tooth movement.
