摘要
根据纤维蛋白原在凝血酶的催化下转化为纤维蛋白的反应原理,建立一种新的凝血酶活性的检测方法。利用紫外扫描仪将此反应的反应物与生成物进行全波长扫描,取二者吸光度的最大差异处波长(250.0nm),在此波长下,绘制不同比活的凝血酶与纤维蛋白原的反应时间与吸光度的变化曲线,根据其变化规律,选择吸光度为0.800时的反应时间为初凝时间,进而拟合出凝血酶活性标准曲线。利用紫外法建立的凝血酶活性检测标准曲线,其线性相关系数(R2)可达0.9987。该法操作快速简便,可广泛应用于凝血酶活性的检测。
This method is based on the reaction mechanism of thrombin fibrinogen change to fibrin which catalyzed by thrombin.scanning the reactant and the product of coagulation on full wavelength by ultraviolet scanner and observing the wavelength,the greatest difference in 250.0nm was selected.At this wavelength,the curves of absorbance changing with the reactant time of different specific activities of thrombin and fibrinogen could be drawn.According to its variation,the duration from reaction beginning to the time of the absorbance reach to 0.800 was selected as the initial setting time.The standard curve of thrombin activity was fitled,which has a linear correlation as high as 0.9987.This method is simple and sensitive,which could be widely used to the detection of thrombin activity.
