摘要
目的:研究三七、白芨、蒲黄、仙鹤草水提物体外止血效果,并进行比较。方法:以内部沸腾法制备上述4种中药水提物。试验分为空白对照(等容生理盐水)组与三七、白芨、蒲黄、仙鹤草水提物(10 mg/ml)组,采用纤维蛋白原平板法测定凝血酶活性以绘制标准曲线,测量反应的溶圈面积及计算凝血酶活性促进率。取家兔血浆随机均分为正常对照(等容生理盐水)组、云南白药(3.3 mg/ml)组与三七、白芨、蒲黄、仙鹤草水提物(3.3 mg/ml)组,测定家兔血浆复钙时间、血小板黏附率、血小板聚集率。结果:与空白对照组比较,三七水提物组溶圈面积增大,凝血酶活性促进率升高;白芨、蒲黄、仙鹤草水提物组溶圈面积减小,凝血酶活性促进率降低,差异均有统计学意义(P<0.01)。与正常对照组比较,三七、白芨、蒲黄水提物组家兔血浆复钙时间缩短,仙鹤草水提物组家兔血浆复钙时间延长;三七、白芨水提物组家兔血小板黏附率、血小板聚集率升高,蒲黄、仙鹤草水提物组家兔血小板黏附率、血小板聚集率降低,差异均有统计学意义(P<0.01)。结论:4种中药水提物中,三七水提物的体外止血效果最显著。
OBJECTIVE:To study and compare in vitro hemostatic effects of the water extract from Panax notoginseng,Rhizoma Bletillae,Typha angustifolia and Agrimonia pilosa. METHODS:Inner ebullition method was adopted to prepare 4 kinds of herbal water extract of mentioned above. In the experiment,there was a blank control group(isovolumic normal saline)and P. notoginseng,R. Bletillae,T. angustifolia and A. pilosa water extract groups(10 mg/ml). Fibrinogen plate method was adopted to determine thrombin activities so as to draw standard curves,measure dissolution circle areas in the reaction and calculate the thrombin activity promoting rates. Plasma of rabbits was randomly divided into normal control group(isovolumic normal saline),Yunnan Baiyao group(3.3 mg/ml) and P. notoginseng,R. Bletillae,T. angustifolia and A. pilosa water extract groups(3.3 mg/ml). The plasma recalcification time,platelet adhesion rates and platelet aggregation rates of the rabbits were determined. RESULTS:Compared with blank control group,dissolution circle areas and the thrombin activity promoting rates in P. notoginseng water extract groups were increased,while those were decreased in R. Bletillae,T. angustifolia and A. pilosa water extract groups,with significant difference(P〈0.01). Compared with normal control group,the plasma recalcification time in groups of P. notoginseng,R.Bletillae and T. angustifolia water extract groups was decreased and increased in A. pilosa water extract group;the platelet adhesion rates and aggregation rates in groups of P. notoginseng and R. bletillae water extract groups were increased and decreased in T. angustifolia and A. pilosa water extract groups,with significant difference(P〈0.01). CONCLUSIONS:Among the 4 kinds of water extract from herbs,the P. notoginseng water extract has the most hemostatic effect in vitro.
