摘要
目的了解普伐他汀对2型糖尿病大鼠视网膜组织中MCP-1表达的影响,探讨普伐他汀对糖尿病视网膜组织中细胞因子的作用。方法将54只Wistar大鼠随机分为正常组、糖尿病组、普伐他汀治疗组,每组18只。糖尿病组和普伐他汀治疗组大鼠用链脲佐菌素诱导建立2型糖尿病模型。于成模后30周取各组大鼠视网膜,分别用免疫组化法和逆转录聚合酶链反应(RT-PCR)法检测视网膜组织中MCP-1的表达水平。结果与正常组大鼠比较,糖尿病组视网膜组织变薄,神经节细胞数目减少;普伐他汀治疗组视网膜形态无明显改变,神经节细胞数目无明显减少。糖尿病组视网膜组织MCP-1表达水平明显高于正常组(P<0.05),普伐他汀治疗组MCP-1表达水平明显低于糖尿病组(P<0.05)。结论普伐他汀能够阻止或延缓糖尿病视网膜形态改变,减弱糖尿病大鼠视网膜组织中MCP-1的表达。
Objective To investigate the effect of pravastatin on the expression of MCP-1 in the retina in rat model of type 2 diabetes and the effect of pravastatin on cytokines in diabetic retina.Methods A total of 54 Wistar rats were randomly divided into normal group,diabetes group,and pravastatin treatment group(n =18 each group).Type 2 diabetes was induced by streptozotocin in rats of diabetic and pravastatin treatment groups.The rats were killed after 30 weeks,and the expression of MCP-1 in the retinal tissue was detected with immunohistochemistry and reverse transcription polymerase chain reaction.Results Compared with normal group,the thickness of the retina became thin and the number of ganglion cells decreased in diabetes group.Such changes were not found in pravastatin treatment group.The expression of MCP-1 in diabetes group was significantly higher than that in normal group(P 0.05),and it was significantly lower in pravastatin treatment group than in diabetes group(P 0.05).Conclusion Pravastatin could prevent and delay the morphological changes in diabetic retinopathy and decrease the expression of MCP-1 in diabetic retina.
